Myszy z niedoborem limfocytów B przez traktowanie króliczymi przeciwciałami anty – mysie IgM ( anty -mu) od urodzenia nie odpowiadają po podaniu antygenów rozpuszczalnych białek w CFA, co zmierzono za pomocą proliferacji limfocytów T po prowokacji antygenem in vitro.
Rolę limfocytów B w pobudzaniu limfocytów T in vivo badano przez adopcyjne przeniesienie limfocytów B swoistych dla haptenu do myszy anty -mu, a następnie immunizację haptenowanym Ag w CFA. Odpowiedź proliferacyjna komórek T na OVA myszy anty -mu BALB/c została częściowo przywrócona przez podanie komórek B specyficznych dla TNP lub FITC i immunizację odpowiednio TNP -OVA lub FITC-OVA.
Ta rekonstytucja była swoista wobec Ag, ponieważ limfocyty B wiążące hapten przywróciły odpowiedzi limfocytów T na OVA u myszy immunizowanych tym samym haptenem sprzężonym z OVA. Mechanizm odtwarzania limfocytów B pobudzania limfocytów T u myszy anty -mu został zbadany przy użyciu transferów limfocytów B rodzicielskich do F1. Wzorzec restrykcji Ia aktywowanych limfocytów T z tych myszy wskazywał, że wystąpiła zarówno bezpośrednia prezentacja Ag przez przeniesione komórki B, jak i za pośrednictwem przeciwciał wzmocnienie prezentacji Ag przez niebędące B, prezentujące Ag komórki gospodarza. Cytokina prozapalna IL-1 beta promuje wzrost guza raka płuc Lewisa poprzez indukcję czynników angiogennych: analiza in vivo interakcji guz-podścielisko.
- Stany zapalne są związane z rozwojem nowotworu. IL-1beta jest wielofunkcyjną i prozapalną cytokiną, która wpływa na prawie wszystkie typy komórek. Aby zbadać rolę IL-1beta we wzroście guza in vivo, transdukowaliśmy wektor retrowirusowy kodujący ludzki gen IL-1beta do mysich komórek raka płuc Lewisa (LLC), a następnie zaszczepiliśmy transformant (LLC/IL-1beta) do syngenicznego C57BL/ 6 myszy.
- Nowotwory wywodzące się z LLC/IL-1beta rosły szybciej (240%, dzień 18, vs null-vector control LLC/neo; p < 0,01) i wykazywały bardziej obfite unaczynienie (250%, vs LLC/neo; p < 0,05), podczas gdy Komórki LLC/IL-1beta, komórki LLC/neo i komórki LLC typu dzikiego nie wykazywały żadnej znaczącej różnicy w szybkości wzrostu in vitro.

- Hybrydyzacja in situ mRNA HGF w skrawkach guza LLC/IL-1beta wykazała, że fibroblasty zrębu i naciekające komórki wykazują nadekspresję mRNA HGF.
- Ponadto, gdy hodowane w obecności HGF in vitro, komórki LLC/IL-1beta wydzielały jeszcze większe ilości czynnika wzrostu śródbłonka naczyniowego i białka zapalnego makrofagów-2.
- Środek antyangiogenny TNP -470 i anty – CXCR2 Ab hamował wzrost nowotworu komórek LLC/IL-1beta in vivo . Wyniki te wskazują, że wydzielanie IL-1beta do środowiska nowotworu indukuje kilka czynników angiogennych z komórek nowotworu i zrębu, a zatem sprzyja wzrostowi nowotworu poprzez hiperneowaskularyzację.
Indukowany haptenem model nieswoistego zapalenia jelit u myszy: odpowiedź immunologiczna błony śluzowej i ochrona przez tolerancję.
- Przedstawiamy tutaj mysi model doświadczalnego przewlekłego zapalenia jelita grubego, w którym podanie kwasu trinitrobenzenosulfonowego (TNBS) w 50% etanolu wywołało zapalenie jelita grubego u podatnych (C3H/HeJ i BALB/c), ale nieodpornych (C57BL/6 i DBA/2 ) szczepy myszy .
- Zapytaliśmy, czy odpowiedzi komórek B specyficzne dla śluzówkowego trinitrofenylu ( TNP ) były indukowane u myszy z zapaleniem okrężnicy indukowanym TNBS i czy indukcja tolerancji na TNBS przez doustne podawanie tego haptenu chroniła myszy przed rozwojem zapalenia okrężnicy. Izotypy i podklasy poliklonalnych i TNP -specyficznych komórek tworzących Ab (AFC) oceniano w śluzówkowych i obwodowych tkankach limfoidalnych myszy C3H/HeJ z zapaleniem okrężnicy wywołanym przez TNBS.
- W zapalnej blaszce właściwej jelita grubego stwierdzono zwiększoną liczbę komórek wydzielających IgA i IgG. Tkanka okrężnicy w stanie zapalnym zawierała również wysokie częstotliwości IgG anty – TNP AFC (głównie podklas IgG1, IgG2a i IgG2b); jednak anty –
- Odpowiedzi TNP w tkankach błony śluzowej bez stanu zapalnego myszy z zapaleniem okrężnicy wykazywały dominujące IgA i IgM z niskimi odpowiedziami IgG anty – TNP . Komórki T CD4+ stymulowane splenocytami TNP wytwarzały więcej IFN-gamma i mniej IL-4, co sugeruje odpowiedź typu Th1.
- Doustne podanie TNBS przed wywołaniem zapalenia okrężnicy znacznie zmniejszyło odpowiedzi śluzówkowe anty – TNP i całkowicie zahamowało odpowiedzi anty – TNP IgG2a i IgG2b.
- Myszy kontrolne nie wykazywały zahamowania odpowiedzi anty – TNP AFC ani zapalenia okrężnicy wywołanego przez TNBS. Uwrażliwienie dookrężnicze podatnych myszy C3H/HeJ z TNBS indukuje zlokalizowaną odpowiedź IgG anty – TNP B w tkance w stanie zapalnym, podczas gdy wcześniejsze doustne podanie TNBS skutkuje brakiem odpowiedzi na ten środek i chroni myszy przed rozwojem indukowanego przez TNBS zapalenia okrężnicy.
Ontogeneza funkcji limfocytów myszy . II. Rozwój zdolności do wytwarzania przeciwciał jest modulowany przez limfocyty T.
- Względną funkcjonalną dojrzałość populacji komórek T i B śledziony noworodków myszy oceniano przez porównanie zdolności do odpowiedzi na antygen niezależny od grasicy , DNP-Ficoll lub SRBC zależny od grasicy przez wytwarzanie przeciwciała in vitro.
- Chociaż mysie komórki śledziony reagowały na DNP-Ficoll we wcześniejszym wieku niż reagowały na SRBC lub TNP -SRBC, przyczyna opóźnienia w odpowiedzi zależnej od T była mylona przez odkrycie dużej liczby limfocytów T supresorowych w śledzionie noworodków . Tak więc niewielka liczba komórek T lub tymocytów śledziony noworodków znacząco zmniejszyła odpowiedź przeciwciał dorosłych komórek śledziony in vitro.
- Chociaż limfocyty B wydają się funkcjonować dojrzałe wkrótce po urodzeniu, ich aktywność może być modulowana przez nadmiar limfocytów T supresorowych; np. przywrócenie funkcji komórek pomocniczych w śledzionie noworodka wymagało leczenia anty -theta przed dodaniem dorosłych komórek pomocniczych. Aktywność hamująca przypisywana limfocytom T wydaje się odgrywać dominującą rolę w określaniu zdolności noworodka do pozytywnej lub negatywnej reakcji na stymulację antygenową
Blokada receptora IL-6 hamuje początek autoimmunologicznej choroby nerek u myszy NZB/W F1.
- W niniejszym badaniu zbadaliśmy prewencyjny wpływ przeciwciała przeciw mysim receptorowi IL – 6 (IL-6R) , MR16-1, na rozwój autoimmunologicznej choroby nerek u samic myszy NZB/W F1 (BWF1). Tolerancję immunologiczną na MR16-1 lub przeciwciało kontrolne o dopasowanym izotypie , KH-5, indukowano przez jednoczesne podawanie anty -CD4 MoAb myszom.
- Następnie myszom podawano dootrzewnowo 0,5 mg MR16-1, 0,5 mg KH-5 lub sól fizjologiczną raz w tygodniu od 13 do 64 tygodnia życia. Leczenie MR16-1 radykalnie stłumiło białkomocz i wydłużyło czas przeżycia myszy BWF1. Tylko jedna na 10 myszy zmarła z wysokim poziomem białkomoczu podczas całego eksperymentu.
- MR16-1 prawie całkowicie tłumił wytwarzanie form IgG przeciwciał anty -DNA i anty – TNP , ale nie form IgM tych przeciwciał . W szczególności, wszystkie podklasy IgG (IgG1, IgG2a, IgG2b i IgG3) wytwarzania przeciwciał anty -DNA zostały znacząco stłumione.
- Co więcej, poziomy IgG1, IgG2a i IgG3 w surowicy u myszy traktowanych MR16-1 były niższe niż u myszy traktowanych solą fizjologiczną i KH-5, podczas gdy poziomy IgM i IgA w surowicy nie uległy zmianie.
